Updated 7 October 2026. Epitalon, also written Epithalon, is the synthetic tetrapeptide Ala-Glu-Asp-Gly (AEDG). It is widely discussed in longevity communities, but claims about reversing ageing, extending human life or restoring telomeres are not supported by modern, confirmatory clinical trials.
What is Epitalon?
Epitalon was developed from research on pineal peptide preparations. Laboratory work has explored possible effects on melatonin-related signalling, oxidative stress, gene expression and telomerase. These are mechanistic observations. They do not establish that the peptide extends lifespan or treats age-related disease in people.
What does the evidence show?
A 2025 scientific review summarized more than two decades of in-vitro, animal and computational research. It described a broad set of proposed biological effects while also emphasizing major gaps in physicochemical, structural and translational knowledge.
Human claims require particular care because older publications frequently studied Epithalamin, a complex pineal extract, rather than the defined four-amino-acid Epitalon molecule. Results for an extract cannot be treated as proof for a synthetic tetrapeptide. Much of the clinical literature is old, regionally concentrated, small and difficult to reproduce against current trial standards.
The 2026 FDA review
In July 2026, the FDA Pharmacy Compounding Advisory Committee considered Epitalon free base and acetate for possible inclusion on the US 503A Bulks List, focusing on a nominated use for insomnia. FDA materials proposed that neither form should be included.
The FDA has stated that it has not identified adequate safety information for the proposed routes and highlighted possible immunogenicity from aggregation and peptide-related impurities. Committee consideration is not an approval and does not validate anti-ageing or sleep claims.
Telomeres: an attractive but incomplete story
Telomerase activity in cells is a laboratory endpoint, not a direct measure of healthier or longer human life. Telomere biology is complex, differs across tissues and also intersects with cancer biology. A credible longevity claim would require long, controlled human trials measuring clinically meaningful outcomes and harms.

- Reproducibility in independent, modern clinical trials
